Intracellular Protein–Drug Interactions Probed by Direct Mass Spectrometry of Cell Lysates

Understanding protein–ligand interactions in a cellular context is an important goal molecular biology and biochemistry, particularly for drug development. Investigators must demonstrate that drugs penetrate cells specifically bind their targets. Towards end, we present native mass spectrometry (MS)-based method analyzing uptake target engagement eukaryotic cells. This based on our previously introduced direct-MS rapid analysis of proteins directly from crude samples. Here, enables label-free studies protein–drug binding human used to determine affinities lead compounds We anticipate this will enable the application MS range problems where important, including protein–protein interactions, binding, characterization therapeutic proteins.
